The PKCzeta expression in prostate cancer and its effect on the proliferation of prostate cancer cell lines

Abstract：Objective To investigate the expression of PKCzeta in prostate cancer and its correlation with the pro liferation of prostate cancer cell lines.Methods The expression of PKCzeta was measured by immunohistochemistry in 19 cases of prostate cancer and 4 cases normal prostate tissues.According to the tumor grade,the prostate tissues were divided into 3 groups.Hilymax was used to transfect the plasmid of PKCzeta in PC3 and DU145 cell lines.The mRNA and protein levels were assessed by RT-QPCR and Western Blot.The ability of cell proliferation was evaluated in PC3 and DU145 cells by CCK-8 kit and plate colony forming.Results The expression of PKCzeta was significantly increased in prostate cancer (P ＜0.01) and had higher level in low tumor grade (Grade 1) (P ＜0.001).The mRNA and protein levels of PKCzeta showed a higher level in DU145 cells,which contributed to a decrease in proliferation.The proliferation ability of PCa cells was inhibited by PKCzeta overexpression.Conclusion A negative correlation exists between PKCzeta expression and tumor grade,which suggested PKCzeta may play a role to protect from further progression of prostate cancer and control the proliferation of prostate cancer cell lines.
