Modulation of imprinted gene CDKN1C on the biological functions of trophoblasts

Abstract:Objective To investigate the effects of imprinted gene,CDKN1C,on the biological functions of human trophoblasts.Methods Biological functions of trophoblasts (TEV-1) were analyzed after CDKN1C silencing by siRNA interference.The mRNA and protein expression of CDKN1C was detected by real-time fluorescent quantitative PCR and Western blot,respectively.Cell proliferation was detected by CCK-8 cellular proliferation assay,cell cycle distribution and apoptosis were detected by flow cytometry,and cell migration and invasion were detected by transwell assay.Results CDKN1C mRNA and protein expression decreased 48 h after siRNA intervention;both early apoptosis rate and late apoptosis rate of siRNA group significantly decreased;and proliferation index significantly increased.Cell cycle assay showed that G0/G1 phases decreased,as S phase and G2/M phases significantly increased.Migration ability and invasion ability of siRNA group were enhanced 48 h after intervention.Conclusion Biological functions of trophoblasts (TEV-1),including proliferation,cell cycle,apoptosis,cell migration and invasion,could be modulate by CDKN1C expression.
