Effects of Ad-HIF-1α-564/402 gene transfer on the heart function in a rat model of acute myocardial infarction

Abstract:Objective To investigate the effect of the recombinant adenovirus containing the 564/402-point-mutant HIF-1 α gene (Ad-HIF-1 α-564/402) on the heart function in the rat model of myocardial infarction.Methods After the accomplishment of acute myocardial infarction model,36 male SD rats were randomly divided into 3 groups,Saline group (n =12),Ad-null group (n =12),and Ad-HIF-1αα-564/402 group (n =12).The ischemic myocardium received saline,Ad-Null,or Ad-HIF-1α-Trip 0.5 mL (1.0 × 108PFU),respectively,by intramuscular injection.Seven days after gene transfection,the cardiac function was examined by echocardiography,the level of creatine kinase MB (CK-MB) isoenzyme and cardiac troponin I (cTnI) in serum were assessed,as well as triphenyl tetrazolium chloride method (TTC) method was performed to evaluate the myocardial infarct size.Results On the 7th day after gene-transfection,the echocardiography revealed obvious better left ventricular ejection fraction (LVEF) and cardiac output (CO) in Ad-HIF-1α-564/402 group than the other two groups (P <0.05);and also better left ventricular fractional shortening (LVFS),but there was no outstanding statistical difference in the three groups (P > 0.05).The levels of CK-MB and cTnI in the Ad-HIF-1αα-564/402 group were significantly the lowest among the three groups (P < 0.05).The myocardial infarct size evaluated by TTC of Ad-HIF-1αα-564/402 was significantly smaller than the other two groups (P < 0.05).There was no obvious difference in CK-MB,cTnI,LVEF,CO,or the size of myocardial infarction between Saline and Ad-Null groups (P > 0.05).Conclusion Ad-HIF-1 αα-564/402 can reduce the degree of myocardial infarction,diminish the infarct size and improve the heart function in rats with ischemic heart in the short term.
