Abstract:Objective To detect the dendritic cell (DC) subsets in peripheral blood and in pleural effusion of patients with tuberculous pleurisy (TP),and to analyze the correlation of DC subsets with the level of IL-12,for exploring its clinical significance. Methods Peripheral blood and pleural effusion from 16 cases of TP patients and 12 cases of healthy subjects (HC group) were collected.Frequencies of DC subsets were analyzed by flow cytometry, while the enzyme linked immunosorbent assay (ELISA) was used to detect IL-12 level in serum and pleural effusion. Results Frequencies of total DC (tDC) and myeloid DC (mDC) in peripheral blood of TP patients were higher than those in HC group, which were obviously decreased after treatment. Moreover,The ratios of tDC and mDC in pleural effusion of patients were higher than those in peripheral blood. However, the frequency of plasmacytoid DC (pDC) in pleural effusion was lower than that in the peripheral blood.Interestingly,IL-12 in pleural effusion and plasma from the TP patients were significantly higher than that in plasma from the HC group, and it is significantly higher in pleural effusion than that in blood in same individual.Correlation analysis found that IL-12 level was positively correlated with tDC and mDC in plasma as well as in pleural effusion. Conclusion DCs,especially mDC,plays an important role in anti-tuberculosis immune in tuberculous pleurisy.
