Inhibition of the migration and invasion of glioblastoma multiforme U87 cells by RNAi silencing PDZ and LIM domain protein 1

Abstract:Objective To investigate the influence on migration and invasion of glioblastoma multiforme U 87 cell line by interfering PDZ and LIM domain protein 1 (PDLIM1) with siRNA.Methods U87 cell line were transfected with PDLIM1 siRNA and negative control siRNA by Lipofectamine 2000.PDLIM1 was detected by real -time PCR at mRNA levels .After the transfection , migration and invasion capacities were detected by scratch test and trans -well experiments . Western blot assay was used to observed the level of protein expression of EMT associated proteins .Results The mRNA level of PDLIM1 in U87 cells that had been transfected PDLIM1-siRNA was significantly reduced (P<0.001).The ca-pacities of migration and invasion of U87 cells was suppressed notably after the knockdown of PDLIM 1 (P<0.01).The protein expression of N-cadherin , β-catenin and Slug , which was related with the EMT , were significantly reduced . Conclusion PDLIM1 has the ability to regulate the protein expression of N -cadherin and β-catenin by adjusting tran-scription factor slug , which is related with the EMT .
