Overexpression of NDPK-A induced ultrastructural changes in mitochondria in the T24 human bladder cancer cell line

Abstract:Objective To investigate the effect of NDPK-A overexpression on the proliferation and ultrastructure of the T24 human bladder cancer cell line.Methods T24 cells were transfected by NDPK-A expressing plasmid with the aid of lipofectamine, followed by screening under the pressure of G418.Expression of NDPK-A protein was detected by immunocytochemistry.Cell proliferation was determined by MTT assay, and ultrastructure of mitochondria was observed under transmission electron microscope.Results Overexpression of NDPK-A not only caused significant inhibition of proliferation in T24 cells, but also led to pycnosis, swelling, cristae deformation and internal cavitation in mitochondria of T24 cells.Conclusion NDPK-A has detrimental effects on mitochondria in the T24 bladder cancer cells.Inhibitory effect on mitochondria metabolism might be one of the mechanisms through which NDPK-A suppresses proliferation of T24 cells.
