Effect of nestin on the anti-apoptosis mechanism of osteosarcoma cells

Abstract:Objective To investigate the effect nestin, a marker of tumor stem cells, on the anti-apoptosis mech-anism of in tumor cells.Methods The osteosarcoma cell line MG63 was utilized for the following investigations.After 0.9μmol/L cisplatin treatment for 12 h and immunohistochemistry ( IHC) staining, the characteristics of surviving cells were observed under the microscope.Damage of 0.9μmol/L cisplatin treatment on nestin-positive cells and nestin-negative cells were compared using comet assay.Results Microscopic observation showed that a small amount of cells survived af-ter cisplatin treatment, the majority of which were nestin-positive.Comet assay demonstrated that damage of cisplatin treatment on nuclear chromosomes in nestin-positive cells was weaker than in nestin-negative cells.Conclusion In the osteosarcoma cell line MG63, nestin may exert a protective role on the nucleus and provide chemotherapy resistance in cisplatin-induced DNA damage.
