Rejuvenation of old mesenchymal stem cells by young cell derived bioactive factors

Abstract:Objective To investigate the effects of human mesenchymal stem cells ( hMSCs ) derived bioactive factors on hMSCs function.Methods Old hMSCs were exposed to a young cell derived condition medium (CM).Con-versely, young hMSCs were exposed to an old cell derived CM .The transwell tests were used to detect the migration capa-bility of hMSCs .The differentiation capability was compared by inducing hMSCs into adipogenic and osteogenic cells .The effect of condition medium bioactive factors on hMSCsenescence were detected by β-galactosidase staining .The changes in microRNA (miRNA) expression were determined by quantitative real -time PCR (qRT-PCR).Results Old hMSCs can be rejuvenated by exposure to a young cell derived CM .The hMSCmigration and differentiation ability was increased , and senescence was decreased .Conversely , the migration and differentiation ability of young hMSCs was impaired by ex-posure to an old cell derived CM .And the senescence was increased .The expression of miR-10a was down-regulated in the old hMSCs derived CM , but was up-regulated in the young hMSCs derived CM .Conclusion Young cell derived bioactive factors can rejuvenate old hMSC function by increasing the expression of miR -10a.
