The experimental study of treatment effect of Interceed on intrauterine adhesions in New Zealand white rabbits

Abstract:Objective To observe the effect and safety of oxidized regenerated cellulose on the treatment of intra -uterine adhesion in New Zealand white rabbits .Methods The intrauterine adhesions rabbit model was established by mechanical injury and infection double -injury method.Forty-eight female rabbits were divided into 4 groups: normal control group , Interceed control group , model group , and Interceed therapy group;in which the rabbits received sham op-eration , Interceed placement , modeling alone or modeling with Interceed treatment , respectively .Interceed placement was performed 1 week after modeling.Each 4 rabbits were sacrificed in each group before treatment (1 week after modeling), and 2 and 4 weeks after treatment .The uterine tissues were collected .HE staining was performed for endometrial morpho-logic observation and gland count .Masson staining was performed to detect the degree of endometrial fibrosis .The expres-sion of TGF-β1 protein was detected by immunohistochemistry .Results (1) There was no significant difference between the normal control group and the Interceed control group in the endometrial gland count , the ratio of fibrosis area or the ex-pression of TGF-β1 protein (P>0.05).(2)Before the treatment, the endometrial gland counts in the model group and Interceed therapy group were significantly lower than that in the normal control group (P<0.001), while the ratio of fi-brosis area and the expression of TGF -β1 protein were significantly higher than those in normal control group ( P<0.001).(3)Two and 4 weeks after treatment, the expression of TGF-β1 protein in the Interceed therapy group was sig-nificantly higher than that in normal control group (P<0.001), while there was no significant difference in the endometri-al gland count or the ratio of fibrosis area between the Interceed therapy group and normal control group (P>0.05).How-ever, there were significant differences in endometrial gland count , the ratio of fibrosis area and the expression of TGF -β1 protein between the Interceed therapy group and the model group (P<0.001).Conclusion The Interceed treatment for intrauterine adhesion is effective and safe , without obvious adverse reactions on the endometrium in New Zealand white&nbsp;rabbit.
