Effects of oxysophocarpine on proliferation and apoptosis of human colon cancer cells

Abstract:Objective To investigate the inhibition of oxysophocarpine (OSC) on proliferation of human colon cancer cell line SW480 and induction on apoptosis , and to explore its mechanism.Methods SW480 cells were treated with OSC (0.5 -2 mg/mL) and the positive control drug 5 -fluorouracil (10 μg/mL) for 12 -48 h.MTT assay was used to detect the proliferation of SW480 cells; fluorescence double staining was used to observe the changes in cell mor -phology and the ratio of apoptotic cells ; flow cytometry was used to demonstrate the presence of apoptosis ; and real -time PCR detection was used to demonstrate changes in gene Caspase -3 and Bcl -2 expression.Results The results of MTT showed that OSC significantly inhibited the growth of SW 480 cells in dose -and time -dependent manners, and morpho-logical characteristics of apoptosis were also observed .The percentages of the G 0 /G1 phase and the S phase cells were in-creased after treated by OSC.Apoptosis and G0 /G1 arrest were induced by OCS in SW480 cells.Apoptotic gene Caspase -3 mRNA was up -regulated, while Bcl -2 mRNA down -regulated.Conclusion OSC inhibits the proliferation of SW480 cells in vitro, involving arrest of cell cycle progression and apoptosis .
