Effects of down-regulation of human angiogenin on the growth of transplanted bladder cancer and the expres-sion of p-Akt, p-GSK3βand p-mTOR

Abstract:Objective To determine the effects of down -regulation of angiogenin on the expression of p -Akt, p-GSK3βand p-mTOR as well as the growth of transplanted bladder cancer tumor .Methods The specific siRNA ANG expression vector and a non -homologous negative control were designed , constructed , and transfected into bladder cancer T24 cells with Lipofectamine 2000.The positive clones were screened by G418, and were named as T24-siANG and T24-NC, respectively.T24 cells without transfection were named as control (T24 group).Tumor cells of the three groups were injected into the back of BALB/c nude mice to establish the xenograft models .Change of micro-blood ves-sels in tumor tissue and expression of CD 31 were detected by Immunohisto -chemical and HE staining .Immunohisto-chemical assay was used to detect the expression of ANG , p-Akt, p-GSK3βand p-mTOR in the tumors.Results BALB/c nude mice receiving ANG siRNA transfected T 24 cells showed significant facilitation of the tumor with lighter tumor weight, lower density of microvessel , and lower CD31, ANG, p-Akt, p-GSK3βand p-mTOR expression than those in the control group .Conclusion Down-regulation of angiogenin inhibits the growth of transplanted bladder cancer in nude mice by down-regulating the expression of p -Akt, p-GSK3βand p-mTOR.
