The correlation between CADM1 expression and promoter methylation in the colorectal cancer cell lines

Abstract：Objective To detect the promoter methylation and expression of CADM 1 gene in colon cancer cells . Methods RT -PCR was used to supervise the CADM1 gene expression level of mRNA .Western blotting was applied to validate the expression of CADM1 protein.BSP and MSP were employed to detect the methylation status of CADM1 gene promoter.Results CADM1 mRNA negative was detection in 24% of colon cancer cell lines , while down -regulated CADM1 mRNA expression was observed in 50% of cell lines.Methylation of CADM1 promoter and CADM1 expression loss were revealed in 75% of colorectal cancer cell lines .Conclusion The methylation of CADM1 promoter is closely related to pathogenesis of colon cancer , providing a probable tumor mark for early screening and prognosis prediction .
