Effects of xenogeneic acellular bone on the astrocyte cells

Abstract:Objective To observe the effects of xenogeneic acellular bone on the proliferation , migration and characteristic of astrocyte cells .Methods The xenogeneic bone was prepared by epoxy crosslinking technology after chemical and physical processing .Different concentrations (0.05, 0.1, 0.2 g/mL) of xenogeneic bone extracted liquid were added into the culture mediums with astrocyte cells .Scanning electron microscope ( SEM) was used for morphologic observation of astrocyte cells .MTT method was used detect the proliferation of the astrocyte cells , and to optimize the con-centration of xenogeneic bone extracted liquid .Transwell method and flow cytometry were used to access the cell migration and cell cycle , respectively .Results The astrocyte cells were polygonal , and were in a good state under SEM .The opti-mized concentration of xenogeneic bone extracted liquid was 0.05 g/mL, which had a positive effect on cell proliferation . In this concentration , the number of transwell cells was increased ( P<0.05) , with no alteration on the cell cycle ( P>0.05).Conclusion Xenogeneic acellular bone has low influence on astrocyte cells , providing good biosecurity to astro-cyte cells.
