Abstract:Objective To study the reversal effect and mechanism of borneol on cisplatin ( DDP) resistance in human esophageal carcinoma cell Eca 109/DDP.Methods The cytotoxic effect of borneol or DDP on Eca 109/DDP cells was measured by MTT and the drug -resistant reversal index (RI) was calculated. The expression of P-glycoprotein (P-gp) was detected by Western blot .Results MTT assay showed that borneol at the concentration no more than 1.560μg/mL had no obvious cytotoxicity to Eca 109/DDP cells ( P>0.05 ) , but it could significantly enhance the cytotoxicity of DDP (1 μg/mL) to Eca109/DDP cells in a concentration -dependent manner ( P <0.05), reduce IC50 of DDP on Eca109/DDP cells (P<0.05), and down-regulate the expression of P -gp of Eca109/DDP cells (P<0.05).RI of DDP was 5.31.Conclusion Borneol possesses a reversal effect on DDP resistance in Eca 109/DDP cells. The mechanism may be associated with the down -regulation of P-glycoprotein .
