The protective effect of fasudial on the myocardial ultra-structure induced by ischemia-reperfusion injury in rat

Abstract：Objective To investigate the protective effect of fasudial on the myocardial ultra -structure induced by ischemia-reperfusion (I/R) injury in rats.Methods Twenty-five Sprague Dawley rats were randomly divided into 4 groups:control group, I/R group, I/R+FL group and I/R+FH group.Using the Langendorff model in isolated working rat heart, after pre-perfusion 20 min, hearts from I/R group were subjected to 45 min of global ischemia followed by 30 min reperfusion.The hearts from I/R+FL group and I/R+FH group were given with perfusate fasudil hydrochloride injec-tion by 10 and 30 mg/kg, respectively, at reperfusion;while 95 min continuous perfusion was given to control hearts .The myocardial ultra-structure and the RhoA , ROCK-2 protein expression were analyzed by transmission electron microsco-py and confocal laser scanning electron microscope , respectively .Results According to electron microscopy analysis , contraction band , mitochondria gathering , arranging in disorder , swelling in individual , different in sizes , with cristae and vacuoles forming, sarcoplasmic reticulum dilatation, and apoptotic morphological changes were observed in I /R group.In I/R+F groups, the damage of myocardial myofilament and mitochondria were significantly reduced , though presenting with cell apoptosis morphological changes .RhoA and ROCK-2 protein expression levels were significantly higher in I/R group and I/R+F group than that in the control group .Conclusion Fasudil has a protective effect on the myocardial ul-trastructure changes induced by ischemia -reperfusion injury .
