The effect of miR-645 on the biological function of osteosarcoma cell line U2OS

Abstract:Objective Investigate the expression of recombinant eukaryotie expression vector of hsa -miR-645 inhibitor and its effects on the proliferation , invasion and the target gene TNFR 2 expression of osteosarcoma cell line U2OS.Methods The pEZX AM02 645 inhibitor was constructed and transfected into U 2OS.The expression of miR-645 and the proliferation of U2OS were verified by RT-PCR and MTT, respectively;while the invasion of U2OS was e-valuated by Transwell .The function of miR-645 was forecasted by bioinformatics tools .The TNFR2 expression in trans-fected U2OS was detected by chemiluminescence method .Results The pEZX AM02 645 inhibitor vector was successfully transfected into U2OS.RT-PCR results indicated that hsa -miR-645 inhibitor was effectively expressed , while MTT results indicated that hsa -miR-645 inhibitor transfection significantly inhibited cell proliferation activity .Transwell re-sults indicated that hsa -miR-645 inhibitor transfection had significant inhibition effect on cell invasion .Reporter gene results showed that TNFR2 was one of the targets of miR -645 in the U2OS cells.Conclusion Eukaryotic expression vector pEZX AM02 645 inhibitor is constructed and effectively expressed in U 2OS cel1s, and TNFR2 is the one of the tar-gets.
