The correlation between MMR gene deficiency and 5-FU sensitivity in colorectal cancer

Abstract:Objective To analyze the correlation between mismatch repair gene ( MMR) expression and chemo-therapeutic drugs 5-fluorouracil (5-FU) sensitivity in colorectal cancer.Methods Four colorectal cancer cell lines SW480, HT29, Lovo and HCT116 were selected and cultured .The 5-FU sensitivity was assessed by MTT , while the cell cycle and apoptosis were observed using flow cytometry (FCM).Results The 5-FU sensitivity was higher in MMR gene silenced cell lines Lovo and HCT 116 than that in complete MMR gene cell lines HT 29, SW480, and MSI -H;among which the highest and lowest sensitivities were observed in the colorectal cancer cell line HCT 116 and MSS colon cancer cell line HT29, respectively.According to cell cycle analysis , significant reduction in S phase cell percentage was observed in 5-FU treated Lovo and HT29 cells ( P<0.05 ) .Conclusion MMR gene silencing significantly increases the 5-FU sensitivity in human colorectal cancer cell lines .Furthermore, 5-FU inhibits the cancer cell proliferation in dose-and time-dependent manners .
