The role of Ero1-Lαin hypertension-induced vascular endothelium injury

Abstract:Objective To investigate the role of endoplasmic oxidoreductin -1-like protein alpha ( Ero1-Lα) in hypertension-induced vein endothelium injury , and further to confirm the vital role of endoplasmic reticulum ( ER) stress in the development of hypertension .Methods Human umbilical vein endothelial cells ( HUVECs) were cultured in hypertension mimic status via culturing with different concentrations of angiotensin Ⅱ( AngⅡ) .MTT was used to investi-gate the apoptosis rate of HUVECs , while Western blot and immunofluorescence histochemistry were applied to assess the expression of Ero1-Lα.Results The apoptosis of HUVECs was observed 24 h after stimulus of Ang Ⅱ.With the AngⅡconcentration rising , increase in the apoptosis rate of HUVECs , and Western blot expression and average immunofluo-rescence optical density of Ero 1-Lαwere revealed .Conclusion Ang Ⅱ can induce apoptosis of HUVECs , in which high expression of Ero 1-Lαparticipates .
