The role of signal transducer and activator of transcription 3 in Th17 cell differentiation in spinal cord injury rat model

Abstract:Objective To investigate the expression of p -STAT3(signal transducer and activator of transcription 3) and Th17 cell related signal molecules in the rat model of spinal cord injury , so as to explore the roles of STAT3 and its related signal molecules in spinal cord injury .Methods A total of 75 adult male SD rats were randomly divided into two groups, control group (n=15) and model group (n=15 each time point).Acute spinal cord injury rat model was estab-lished by the modified Allen weight drop method in the model group , while laminectomy alone was performed in control group.Each 15 rats in model group were sacrificed at the 1st, 24th, 48th and 72nd hour after operation for collection of spi-nal cord tissues, spleen tissues, and peripheral blood.Immunohistochemistry method was used to detect the expressions of interleukin ( IL)-17 in damaged spinal cord tissues , Western blot was applied to assess the protein expression of p -STAT3 in the spinal cord and spleen tissues , and ELISA was used to measure serum IL -6, IL-21 and IL-23.Results Expression of IL-17, p-STAT3, IL-6, IL-21 and IL-23 in model group was significantly higher than control group (P<0.05).Furthermore, the expression of p -STAT3 was significantly positively correlated with IL -17, IL-6 and IL-21 (P<0.05).Conclusion Aberrant activation of STAT3 may be one of factors to contribute to Th17 polariza-tion after spinal cord injury .
