Screening of Vaterinary Drug Residues in Animal-derived Food Origin by High Resolution Mass Spectrometry

Abstract:This paper describes the development of mass spectrometry library and the method of screening and confirming 83 drugs and their metabolites in black-bone chicken meat,eggs,beef,and mutton using ultra-high performance liquid chromatography-quadrupole-obitrap high resolution mass spectrometry(UHPLC-Q/Obitrap HRMS).A two-step procedure was employed:homogenized samples were first extracted with 2 mL aqueous solution,followed by a second extraction using 8 mL of acetonitrile.The extract was then passed through a purification column,reconstituted after nitrogen blow-down,and analyzed by UHPLC-Q/Obitrap.The screening detection limits ranged from 5.0-20 μg/kg.Average recoveries across three spiked concentration levels were 60%-110%.This method proved to be straightforward,rapid and reliable,making it suitable for the rapid screening and confirmation analysis of veterinary drug residues in black-bone chicken meat,eggs,beef and mutton.
