Variation analysis of porcine reproductive and respiratory syndrome virus strains isolated from Hebei province in 2010
Abstract:To investigate the variation of porcine reproductive and respiratory syndrome virus in Hebei province, samples collected from six intensive pig farms suffered from PRRSV in 2010 were used for RT-PCR. The ORF5 and Nsp2 genes were subjected to PCR amplification, cloning and sequencing. Sequence analysis showed that the amplified Nsp2 genes of samples shared 91.4%-98.8% amino acid homology and shared 61.4% -99.4 % amino acid homology with reference strains by analysis with soRware DNAStar. The amplified ORF5 genes shared 95%- 99.5% amino acid homology to each other and shared 84.6%-88.6 % amino acid homology with reference strains VR-2332 and 84.1%-87.6% amino acid homology with reference strain MLV. The results showed the six Nsp2 genes amplified from samples collected from 2010 exhibited 30 amino acid deletion (amino acid 482 and amino acid 533 to 561). Phylogenetic analyses showed the six PRRS viruses belonged to North American genotype and could be clustered into the same subgroup. These viruses were closely related to strains SY0608, JXA1, HUN4, WUH1 and HB-3 (cz), but far from strains VR-2332, CC-1 and MLV. The results suggested that porcine reproductive and respiratory syndrome virus collected from intensive pig farms in Hebei province had experienced great variation and the PRRS viruses isolated in 2010 were still highly pathogenic.
