Screening of PIPK-FAB Associated Proteins in Rice and Their Responses to Multiple Environmental Stress Factors

Abstract:In order to further analyze the molecular mechanism conducted by rice phosphatidylinositol phosphate kinase PIPK-FAB in regulating stress responses,the yeast two-hybrid technique was used to screen its interacting proteins(OsPAPs)and combined with the transcriptome analysis,the response characteristics of OsPAPs genes to environmental stresses were analyzed.The yeast two-hybrid assay showed that PIPK-FAB gene was non-toxic to yeast strain AH109 and did not exhibit self-activation activity.A total of 34 PIPK-FAB interaction positive clones were screened from the rice cDNA library.After sequencing,20 interacting proteins were identified,including phosphoinositide-specific phospholipases,SNARE proteins,GTPase-activating proteins,ascorbate peroxidases,calmodulin-like proteins,and lectins.The results of transcriptome analysis showed that OsPAPs genes could respond to various abiotic and biotic stresses such as salinity,drought,temperature extremes,and rice blast infection,showing diversified expression patterns.In summary,PIPK-FAB gene might serve as a key regulatory node in the response network of rice to environmental stress,and cooperate with multiple signaling pathways to participate in the regulation of stress resistance.
