Study on in vitro transcription and translation of the precore variant of hepatitis B virus
Abstract:Objective To investigate the effect of T1862 and A1899 codonmutants of hepatitis B virus (HBV) on the synthesis and excretion of the precursor of hepatitis B e antigen. Methods The entire precore and core region were amplified from the sera of a HBeAg -negative patient with fulminant hepatitis B by polymerase chain reaction (PCR). PCR products were cloned into plasmid pUC18 and sequenced for analysis of precore mutations.Precore and core sequence of T1862/A1899 variant were also cloned into expression plasmid pGEMT for in vitro transcription and translation study on synthesis and excretion of e antigen. Results T1862 /A1899 variant did not interrupt the synthesis of the precursor of e antigen. Conclusion The absence of e antigen in patients with the fulminant hepatitis may be explained by precore codon mutation T1862/A1899, which may exercise its effect on the excretion of the precursor of e antigen, other than the synthesis of precursor protein.
