Abstract:Objective To study the role of bystander effect in the cytocidal activity of TK/GCV against tumor cells. Methods Human pulmonary carcinomal cell line and TK gene-transfected A459 cells were cultured together in varied ratio at low and high cell density respectively, treated with 50 mol/L GCV for 5 d. The living cells were counted by MTT assay. Results When the cells were cultured at a high density (104/well) with only 20% TK+cells, significantly higher growth inhibition than that of control cells (P<0.05) was observed. Obvious growth inhibition was observed (P<0.05) in the cell culture at a low density (103/well) only when TK+cell ratio was raised to 50%. Conclusion Bystander effect plays an important role in the cytocidal activity of TK/GCV against tumor cells, and is quite relative to intercellular contact.
