Correlation between serum lncRNA UCA1,miR-331-3p and the severity and prognosis of severe infection patients

Abstract:Objective To investigate the expression of serum lncRNA UCA1 and miR-331-3p in severe infections and their relationship with the severity of the condition and prognosis.Methods A total of 84 critically infected patients admitted to the Jianli People's Hospital from January 2023 to January 2024 were selected,including 39 males and 45 females,aged(49.8±8.1)years old,ranging from 43 to 59 years old.Patients were divided into sepsis group(n=47)and septic shock group(n=37)based on the severity of severe infection.Another 40 healthy individuals who underwent physical examinations at the People's Hospital of Jianli City,Hubei Province during the same period were selected as the health group,with 19 males and 21 females,aged(51.0±8.7)years old,ranging from 42 to 60 years old.Collect general information(age,gender,body mass index,comorbidities,site of infection)and clinical data[C-reactive protein(CPR),white blood cell count,platelet count,acute physiology and chronic health evaluation Ⅱ(APACHEⅡ)score]of all participants,and record the survival status of severely infected patients one month later as prognosis.Detect the expression levels of serum lncRNA UCA1 and miR-331-3p in the enrolled individuals,predict the interaction between serum lncRNA UCA1 and miR-331-3p,and further study the relationship between various indicators and prognosis in critically infected patients.Results Bioinformatics prediction revealed that there is a targeted binding site between serum lncRNA UCA1 and miR-331-3p.One month later,54 critically infected patients survived and 30 died.Logistic regression analysis showed that APACHEⅡ score and elevated lncRNA UCA1 were independent risk factors for patient prognosis,while elevated miR-331-3p was an independent protective factor for patient prognosis(P<0.05).The results of the subject operating characteristic curve showed that the area under the curve for predicting the prognosis of critically infected patients by combining serum lncRNA UCA1 and miR-331-3p was higher than that predicted by serum lncRNA UCA1 and miR-331-3p alone(P<0.05).Conclusion The expression levels of serum lncRNA UCA1 and miR-331-3p can be used to determine the severity of severe infections,and the combined prediction effect of the two is better than predicting them separately.
