Effect of insulin-like growth factor-1 on vascular remodeling and apoptosis of cardiomyocytes in rats with coronary heart disease

Abstract:Objective To investigate and analyze the effects of insulin-like growth factor-1 (IGF-1) on myocardial cell apoptosis and vascular remodeling in rats with coronary heart disease. Methods 308-week-old SD male rats were selected and randomly divided into control group,model group and IGF-1 group with 10 rats in each group. The model group and IGF-1 group rats were fed with high-fat diet for 7 weeks to construct a rat coronary heart disease (CHD) model. After successful modeling,rats in the IGF-1 group were treated with IGF-1 at a dose of 30 μg/kg,while rats in the control group and model group were treated with an equal amount of physiological saline for 4 consecutive weeks. The cardiac function and coronary artery blood flow of each group of rats were measured before and after intervention. After the intervention,the rats were euthanized and their coronary arteries and left ventricular tissues were taken. HE staining was used to observe the overall specimen situation,and Western Blot experiments were used to detect the expression of apoptosis related proteins such as PI3K,AKT,Bax,and Bcl-2. Results Compared with the blank control group,LVSP,-dp/dt-max,+dp/dtmax and coronary blood flow in model group and IGF-1 group were significantly decreased,the intima of coronary artery was significantly thickened,and the expression levels of ICAM-1 mRNA and VCAM-1 mRNA were significantly increased. Moreover,LVSP,-dp/dtmax,+dp/dtmax and coronary blood flow in IGF-1 group were significantly higher than those in model group,while the intima thickness of coronary artery,ICAM-1 mRNA and VCAM-1 mRNA expression levels were significantly lower than those in model group,with statistical significance(P<0.05). Compared with the blank control group,the expression level of Bax protein in myocardial tissue of model group and IGF-1 group was significantly increased,the expression level of PI3K and Bcl-2 protein was significantly decreased,and the expression level of Bax protein in IGF-1 group was significantly lower than that in model group. The expression levels of PI3K and Bcl-2 protein in IGF-1 group were significantly higher than those in model group,with statistical signifi-cance(P<0.05).Conclusion IGF-1 can inhibit cardiomyocyte apoptosis,reduce cardiomyocyte inflammation,inhibit intima thick-ening and vascular remodeling in coronary heart disease model rats.
