Effect of circular fibroblast growth factor receptor 1/miR-217/HOXA13 axis on colon adenocarcinoma progression and tumor immune invasion

Abstract:Objective By constructing the potential hsa_circRNA/miRNA/mRNA regulatory axis to clarify the potential underlying mechanism of action of circular RNA in colon adenocarcinoma(COAD).Methods Candidate mirnas downstream regulated by Circ FGFR1 were obtained by CRI database,and the final candidate mirnas were identified by expression analysis and prognostic analysis in TCGA database.The downstream target genes of miRNA were predicted in the miRBD database,and the final candidate downstream target genes were identified in the TCGA database combined with expression analysis and prognostic analysis.Multivariate analysis was carried out by Cox regression method,on the basis of which a nomogram was constructed for internal verification.The correlation be-tween target genes and immune cells was analyzed in TIMER database and TCGA database.The possible mechanism of action was in-vestigated by gene set enrichment analysis(GSEA)enrichment analysis.Results HOXA13 was selected as the final candidate down-stream target gene in the CRI database,and COAD patients with high HOXA13 expression had better overall survival(OS).The C-in-dex of the column chart is 0.739(0.712~0.765),and the prediction model has good accuracy.The Timer database showed that HOXA13 expression had a good correlation with all kinds of immune cells.GSEA analysis revealed that HOXA13 may influence COAD by participating in biological events that affect DNA methylation,cell cycle,mitochondrial division,and cholesterol production.Con-clusion The increased expression of HOXA13 can prolong the OS in COAD patients and can be used as a novel predictive biomarker in COAD patients.Meanwhile,HOxA13 may be associated with the tumor immune infiltration of COAD and may play an important in-hibitory role in the immune escape of tumor cells in the COAD tumor microenvironment.The circFGFR1/miR-217/HOXA13 axis may inhibit the progression of colon cancer by influencing the cell cycle,and these findings suggest a potential target and mechanism for the development of COAD anti-cancer strategies.
