Effect of neutralizing antibody of high mobility group box protein-1 on intestinal barrier function in late resuscitation rats with hemorrhagic shock

Abstract:Objective To investigate the effect of neutralizing antibody of high mobility group box protein-1(HMGB1)on intesti-nal barrier function in late resuscitation rats with hemorrhagic shock.Methods A total of 153 Wistar rats were randomly divided into Sham group(n=17),normal temperature resuscitation group(n=34),low temperature resuscitation group and hemorrhagic shock group(n=68),low temperature resuscitation +HMGB1 neutralizing antibody group(HR+anti-HMGB1,n=34).Hemorrhagic shock model was constructed,and intestinal mucosa,mesenteric lymph nodes and blood were collected.Western blot was used to detect the expression of HMGB1 protein in intestinal mucosa of rats in both groups.The expression of serum inflammatory factors was detected by enzyme-linked immunosorbent assay.Hematoxylin-eosin(HE)staining was used to observe the intestinal mucosal injury in the early and late stages of resuscitation in each group.Results The previous study detected the serum HMGB1 expression of hemorrhagic shock rats at 0,2,4,8 and 24 hours after resuscitation,and found that the HMGB1 expression level in HR group gradually increased with the delay of resuscitation time.After treatment with HMGB1 neutralizing antibody,HMGB1 protein expression was down-reg-ulated in HR+ Anti-HMGB1 group,and HMGB1,tumor necrosis factor-α(TNF-α)、interleukin(IL)-1β、TNF-α,IL-1β,IL-6 and IL-8 expression were also down-regulated in peripheral blood,with statistical significance(P<0.05).HE staining results indicated that compared with the HR group after 24 hours of resuscitation,intestinal mucosal injury was aggravated.After 24 hours of resuscitation,the intestinal mucosal damage was not obvious after the administration of HMGB1 neutralizing antibody,which was ba-sically consistent with the early stage of the HR group.Conclusion Intestinal injury is aggravated in lateinducible hypothermia,and inhibition of HMGB1 release can protect the injury caused by late resuscitation inducible hypothermia.
