Molecular mechanism of extracorporeal membrane oxygenation on the expression of cytokines in acute lung injury induced by impact injury

Abstract:Objective To investigate the effects of extracorporeal membrane oxygenation ( ECMO) on acute lung inju-ry. Methods Forty dogs were randomly divided into four groups:Group A,the control group;Group B,acute lung injury group;Group C:mechanical ventilation group;Group D:ECMO group. The lungs were removed under the aseptic condition from dogs of the four groups,and Western blot,Real-Time PCR,Pathological changes of lung tissue for HE staining were ob-served. Results Pro-inflammatory cytokines of IL-1b,IL-4,IL-6 in Group B increased when compared with those in Group A. After mechanical ventilation and ECMO treatment,pro-inflammatory cytokines decreased(P<0. 05). Inflammatory path-ways factor NF-κB expression also significantly decreased. Description of acute lung injury caused by inflammation might be associated with NF-κBpathway. There were no statistically significant difference among Caspase3, Bax and Bcl-2 ( P >0. 01). Conclusion ECMO can improve lung function effectively on acute lung injury.
