Effect of extracorporeal membrane oxygenation on the expression of cytokines in a-cute lung injury induced by impact injury

Abstract:Objective To investigate the specific cytokines in the acute lung injury caused by severe trauma with the treatment of extracorporeal membrane oxygenation(ECMO). Methods Twenty beagle dogs were randomly divided into 4 groups:normal control group,model group,mechanical ventilation treatment group,and ECMO treatment group. Rubber bul-lets hit to the canine model of lung injury. To extract total protein of the lung tissue,proteinchip assayed and screened differ-entially expressed cytokines. Results In the detection of the 20 cytokines,compared with the control group,IL-2,IL-6,IL-8,IL-10,MCP-1,SCF,HGF and HGF R,IL-1B,MIP-1βand TNF RI B of model group,expressed increasingly,which de-creased after mechanical ventilation and ECMO treatment,and the difference of ECMO treatment was more obvious. The ex-pression tendency of VEGF,FGF-7 and IL-12p40 were opposite. The expression of GM-CSF,TNF alpha and IL-17A had no significant difference. EPO and IFNγhad no expression. Conclusion ECMO treatment can improve the inflammatory re-sponse caused by severe trauma induced acute lung injury,and provide direction and target for the subsequent study of mo-lecular mechanism of the acute lung injury caused by severe trauma with the treatment of ECMO.
