Pathogenesis of traumatic pancreatitis via apoptosis-based pancreatic transcriptomic analysis

Abstract:Objective By conducting high-throughput sequencing analysis on the pancreatic tissue of ex-perimental rats with high-altitude traumatic pancreatitis(HTP),this study aims to explore the key differentially ex-pressed genes(DEGs)and related pathological signaling pathways associated with apoptosis after HTP.Methods A total of 18 Sprague Dawley(SD)rats were divided into three groups:control(C)group,high altitude(H)group(rats were kept in a high altitude chamber for 8 weeks,with the altitude set at 5,000 m,air pressure of 47-48 kPa,temperature of 25℃,and oxygen concentration of 10%-11%),and the HTP group(rats were kept in the same high altitude chamber for 8 weeks and then the pancreas was shocked once with a small animal multifunctional shock de-vice,impact pressure=400 kPa).After 24 h of modeling,high-throughput sequencing was performed on pancreatic tissues of all rates,to analyze differentially expressed genes between groups C and H,as well as between groups H and HTP.The intersection genes,identified after combination with apoptosis-related genes,further experienced gene ontology(GO)and Kyoto encyclopedia of genes and genomes(KEGG)enrichment analysis.Then the least absolute shrinkage and selection operator(LASSO)regression analysis was conducted to screen for key genes involved in the occurrence of HTP.Finally,quantitative real-time polymerase chain reaction(RT-qPCR)analysis of pancreatic tis-sues from H and HTP groups was done to verify the results of sequencing data analysis.Results Differential ex-pression analysis revealed 2,029 DEGs between groups C and H and 1,569 DEGs between groups H and HTP.Crossing the two parts of DEGs with apoptosis-related genes revealed 29 intersecting genes.KEGG of these intersec-ting genes was mainly enriched in pancreatic endocrine metabolic pathways related to diabetes and insulin secretion,such as diabetes in late adolescence,type Ⅱ diabetes,and insulin secretion;while GO was mainly enriched in bio-logical processes such as positive regulation of cell secretion,cell response to peptide hormone stimulation,insulin receptor signal pathway,and regulation of peptide hormone secretion.LASSO regression analysis selected three key genes,i.e.,Il33,Ybx1,and Insrr,two of which(Ybx1,Insrr)were validated by PT-qPCR analysis on expression trends.Conclusion The key signal pathways of the HTP course are the pancreatic endocrine metabolism signal pathways related to diabetes and insulin secretion,such as maturity-onset diabetes of the young,type Ⅱ diabetes mel-litus and insulin secretion.The key genes are Ybx1 and Insrr.
