Diagnostic exploration of ferroptosis-related genes in sepsis

Abstract:Objective The aim of this study is to investigate the diagnostic value of ferroptosis-related genes in adult sepsis.Methods We screened and got the microarray datasets of adult sepsis from the GEO database.The training set is GSE 236713(125 patients with sepsis,30 healthy volunteers).The validation set(No.1)is GSE 131761(81 patients with sepsis,15 healthy volunteers).The validation set(No.2)is GSE 95233(51 patients with sepsis,22 healthy volunteers).The training set was used to identify the ferroptosis-related differentially expressed genes(DEGs).Enrichment analysis of the ferroptosis-related DEGs was performed.Hub genes were identified by protein-protein interaction(PPI)networks.The diagnostic value of hub genes in adult sepsis was tested in the validation sets.Results A total of 1 803 DEGs were obtained in GSE 236713,of which 41 were ferroptosis-related DEGs.Enrichment analysis revealed significant enrichment of the hypoxic pathway.Through PPI networks,TP53,PPARG,TLR4,HMOX1,TIMP1 and LCN2 were selected as hub genes.In GSE 131761 and GSE 95233,the AUC of PPARG,TLR4,TIMP1 and LCN2 for the diagnosis of sepsis was greater than 0.91.Conclusion PPARG,TLR4,TIMP1 and LCN2 have potential value in the diagnosis of sepsis in adults.
