Current status and strategies in the development of enterovirus vaccines

Abstract:Hand-foot-mouth disease(HFMD)primarily infects children under 5 years old,causing acute infection,some of which may develop more serious or even life-threatening symptoms.It is one of the most prevalent infectious diseases in China.In 2015,the only HFMD vaccine,an inactivated EV-A71 vaccine,was developed and introduced in China,leading to effective control of HFMD caused by EV-A71.However,this vaccine does not provide protection against other enterovirus serotypes,such as CVA16,CVA6,CVA10 and others.As a result,there has been an increase in the incidence of HFMD caused by these other serotypes.Therefore,the development of multivalent enterovirus vaccine is essential.This paper summaries the pathogenicity of enteroviruses,progress in vaccine development,and the challenges faced by these vaccines.In developing a multivalent enterovirus vaccine,balancing immunodominance among different immunogen is critial,and eliciting both humoral and cellular immunity simultaneously remains a significant challenge.
