Abstract:Methyl-CpG-binding domain (MBD) protein family plays an important role in transcriptional regulation of genes. MBD protein binding function is achieved through functional MBD region and methylated CpG, and results in gene suppression through heterochromatin formation, histone acetylation and methylation of DNA. However, some MBD proteins also bind to non-methylated DNA, and exert transcriptional activation and multiple differentiation potentials through other regulatory domains or interaction with the nucleosome remodeling deacetylase (NuRD)/Mi-2 complex. The gene mutation or abnormal expression of MBD protein can cause many diseases, including nervous system diseases, cancer and chronic hepatitis B. In this study, the main function of the chief members in MBD protein family and their relationship with diseases were summarized.
