Rooting of Pemphis acidula under Sugar-Free Tissue Culture Conditions and Red Mangrove Conservation Research
Abstract:To study the most suitable Ca2+ and Mg2+ concentrations for rooting of red mangroves under sugar-free tissue culture conditions, a comparative experimental study was conducted. It was found that under the conditions of vermiculite, rice husk charcoal, and culture solution ratio of 3:1:4.8, CaCl2·2H2O concentration of 1200 mg/L, and MgSO4·7H2O concentration of 2500 mg/L, the average root length of seedlings was 16.55 cm, the stem diameter was 4.98 mm, the maximum root length was 21 cm, with zero pollution and a transplant survival rate as high as 98%. Therefore, by significantly increasing the concentrations of Ca2+ and Mg2+ in plant tissue culture medium, it is possible to effectively promote the establishment of new root systems in red mangrove seedlings, improve survival rates, and promote their rapid growth. This finding has significant implications for optimizing red mangrove tissue culture techniques and improving seedling production efficiency, and also provides a useful reference for further exploration of optimal conditions for plant tissue culture.
