Association Analysis of Polymorphisms in the FSHβ, ESR, and OPN Genes with Reproductive Traits in Large White Pigs
Abstract:This study used PCR and PCR-RFLP techniques to detect the effects of polymorphisms in the follicle-stimulating hormone β (FSHβ), estrogen receptor (ESR), and osteopontin (OPN) genes on reproductive traits such as litter size, number of live piglets, stillbirths, and mummified fetuses in Large White pigs. The results showed that all three genes exhibited polymorphism (AA, AB, and BB). The dominant allele for the FSHβ gene was B, while for ESR and OPN genes it was A. The pig populations of FSHβ and ESR genes conformed to Hardy-Weinberg equilibrium, whereas the OPN gene population deviated from it. In the FSHβ gene population, there were significant differences (P < 0.05) in the number of mummified fetuses among different genotypes, with AA genotype pigs having the highest litter size and the lowest number of live piglets. In the ESR gene population, there were significant differences (P < 0.05) in litter size among different genotypes, with the order of sow genotypes from highest to lowest being AB > AA > BB. In the OPN gene population, no significant differences (P > 0.05) were found in reproductive traits among different genotypes, but the order of sow genotypes in terms of litter size and number of live piglets was BB > AA > AB. Combined genotype and reproductive trait correlation analysis showed that the optimal molecular marker combination for FSHβ and ESR combined genotypes was AAAB, for FSHβ and OPN was BBBB, and for ESR and OPN was ABBB. Polymorphisms in the FSHβ, ESR, and OPN genes are associated with reproductive traits in Large White pigs and can serve as candidate genes for molecular breeding of Large White pigs.
